A blood sample travels from the collection point to the testing laboratory through pressure changes, temperature fluctuations, and transport vibration. Under the frameworks of IATA PI650, ADR P650, and DOT 49 CFR 173.199, what often determines whether it arrives “compliantly” is not the outermost box, but the transparent bag in the middle marked “95kPa.”
UN3373 refers to “Biological Substance, Category B” — infectious substances that do not carry the same lethal risk as Category A but are still infectious and fall under Class 6.2 dangerous goods. Typical examples include diagnostic blood samples, tissue specimens, urine, nasal swabs, and other human or animal body fluids. In air transport, these are governed by IATA PI650; in European road transport, by ADR P650; and in the United States, by DOT 49 CFR 173.199.
The key point is that compliance for Category B is not a “recommendation” — it is mandatory. If packaging does not meet the standard, carriers can refuse the shipment and regulators can pursue accountability.
The core requirement of 95kPa is that the primary receptacle or secondary packaging must withstand an internal pressure differential of no less than 95 kPa (about 0.95 bar) without leakage, and this performance must hold across a temperature range of −40°C to +55°C.
This figure is not arbitrary. It simulates the actual pressure load on packaging during air transport, when cargo compartment pressurization changes and ground-to-altitude temperature differences combine. A bag that “looks sturdy” does not have a declarable 95kPa capability unless it has been verified by an independent third-party laboratory according to P650 procedures. In other words, the basis for compliance is a test report, not how it feels in your hand.
Compliant UN3373 packaging involves four functional layers, often referred to in the industry as a “triple packaging system”:
Primary Receptacle: the tube or bottle that directly contacts the specimen; it must be leakproof, and liquid samples must not exceed 1 L per primary receptacle;
Absorbent Material: placed between the primary receptacle and the secondary packaging; its absorption capacity must be sufficient to absorb the entire liquid content of all primary receptacles;
Secondary Packaging: the 95kPa biohazard bag, which provides the leakproof barrier and the 95kPa pressure barrier;
Rigid Outer Packaging: must pass a 1.2 m drop test, with total liquid content per outer package not exceeding 4 L.
The 95kPa bag sits at the second layer. Its role is to contain leakage before it reaches the outer packaging if the primary receptacle fails. It must also carry the biohazard symbol and the words “BIOLOGICAL SUBSTANCE, CATEGORY B,” and provide a carrier for the UN3373 diamond mark.
Inside the 95kPa bag, the Absorbent Tube Holder solves a specific problem: tubes move and collide during transport, and if one breaks, liquid spreads inside the bag. The absorbent tube holder uses compartmentalized fixation to keep tubes separated from one another and cushion vibration, while using a stated absorption capacity per compartment (a common specification is 14 mL per compartment) to contain leaked liquid and keep contamination inside the secondary packaging.
It does not replace the 95kPa bag, nor does it replace absorbent material. Instead, it allows “fixation + cushioning + absorption” to function as an operational whole inside the secondary packaging. For multi-tube specimen transport — especially blood collection tubes and viral transport tubes — the absence of this step is often the most common breakpoint in the packaging chain.
Request third-party 95kPa test reports and confirm that the tested temperatures cover −40°C and +55°C, not just room temperature;
Match the tube holder to the tube diameter to avoid the hidden risk of “compliant bag, loose contents”;
Confirm labeling completeness: the biohazard symbol, the UN3373 diamond mark position, and a separate document pouch — all three are indispensable.
